This is a ready-to-use SOP for preparing, checking, approving, and maintaining the master production and control record (MPCR), the approved template from which every executed batch record is generated. Replace every <<FILL: ...>> placeholder with your own specifics, set your document numbers and dates, and route it through your normal document control, review, and approval. A worked filled specimen follows. Verify each cited regulation against the current source before you rely on it.

| Field | Entry |
|---|---|
| Document title | Master Production and Control Record Preparation, Review, and Approval |
| Document number | <<FILL: SOP-ID, e.g. SOP-QA-041>> |
| Version | <<FILL: version>> |
| Effective date | <<FILL>> |
| Supersedes | <<FILL: prior version or "New">> |
| Document owner | <<FILL: role, e.g. Head of Quality Assurance>> |
| Applies to | <<FILL: sites / product lines in scope>> |
1. Purpose
This procedure defines how <<FILL: COMPANY NAME>> creates, checks, approves, revises, and periodically reviews the master production and control record (MPCR) for each drug product, so that every executed batch record it generates is complete, accurate, and traceable to a single approved source.
2. Scope
This procedure applies to the MPCR for every commercial and, where applicable, clinical drug product manufactured, packaged, or tested at the sites listed in the header. It covers initial creation, revision through change control, periodic review, and the relationship between the master record and the executed batch production and control record (BPCR). It does not cover the disposition decision for an individual batch, which is governed by <<FILL: SOP-ID for batch disposition>>.
3. Responsibilities
| Role | Responsibility |
|---|---|
| Preparer (Manufacturing, Process Engineering, or Technical Services) | Drafts or revises the MPCR from the validated process, development report, or approved application, and signs and dates as preparer. |
| Independent checker | Reviews the draft MPCR against the process it represents, confirms completeness and internal consistency, and signs and dates independently of the preparer. |
| Quality Assurance | Approves the MPCR, confirms alignment with the approved application and validation status, and maintains the effective, controlled version. |
| Document control | Issues and retrieves controlled copies, tracks the current effective version, and archives superseded versions. |
| Site / product owner | Confirms the periodic review is scheduled and completed, and initiates change control when the process, specification, or regulatory basis changes. |
4. Definitions
- Master production and control record (MPCR): the approved template for manufacturing a specific product, one per product per site, from which every batch production and control record (BPCR) is reproduced. See 21 CFR 211.186.
- Batch production and control record (BPCR): the executed, batch-specific record generated from the current effective MPCR. See 21 CFR 211.188.
- Independent check: a review performed by a person other than the preparer, confirming accuracy and completeness before the document moves to quality unit approval.
- Theoretical yield: the quantity that would be produced at any appropriate phase based on the quantity of components used, in the absence of loss or error, as defined at 21 CFR 210.3.
5. Procedure
5.1 Draft or revise the MPCR
- Base the draft on the current validated process, the process description in the approved application, and the current specifications for components, in-process materials, and the finished product.
- Include, at minimum, the elements 21 CFR 211.186 requires: product name and strength; dosage form; a complete list of components by name and amount, with a statement of theoretical weight or measure; a description of containers and closures; complete manufacturing and control instructions; sampling and testing procedures and specifications; special notations and precautions; and space for the required signatures at each step.
- Identify every step that requires a second-person verification (for example component charge-in per 211.101, yield calculation per 211.103, line clearance per 211.130) and build the corresponding sign-off field directly into the record.
- State in-process specifications derived from process data, not set independent of the validated process.
5.2 Independent check
- A person other than the preparer reviews the draft against the validated process, the application, and current specifications.
- Confirm internal consistency: components in the formula section match components referenced in manufacturing instructions; in-process limits match the specification document; equipment identified matches what is qualified for the process.
- Sign and date as independent checker. Route back to the preparer for correction if the check identifies a gap; re-check after correction.
5.3 Quality unit approval
- Quality Assurance reviews the checked draft for regulatory completeness (the 211.186 elements), alignment with the approved application, and consistency with the current validation status.
- Approve and assign the document an effective date and version number, or return with documented comments.
- On approval, document control issues the new version as the sole effective MPCR for the product, and withdraws the prior version from active use.
5.4 Generate batch records from the current MPCR
- Reproduce the batch production and control record only from the current effective MPCR; do not carry forward a batch record generated from a superseded version.
- Confirm at batch record issuance that the version number on the issued BPCR matches the current effective MPCR version.
5.5 Periodic review
- Review each MPCR at a defined frequency (
<<FILL: interval, e.g. every 2 years or per the site document control SOP>>) or upon a trigger event (a process change, a specification change, a recurring deviation pattern traced to the master record, or a regulatory commitment change). - Document the review outcome: confirmed current with no change, or routed to change control for revision.
5.6 Revise through change control
- Any change to the MPCR, including a correction, goes through
<<FILL: SOP-ID for change control>>before the revised version becomes effective. - Repeat the preparation, independent check, and quality unit approval steps above for the revised version.
- Record the change in the MPCR’s revision history (section 9) with a summary sufficient to understand what changed and why.
6. Acceptance criteria
- The MPCR contains every element 21 CFR 211.186 requires, with the preparer and independent-checker signatures both present and dated.
- Only one version of the MPCR is effective for a given product at any time, and document control can demonstrate this.
- Every batch record issued was generated from the current effective MPCR version, verified at issuance.
- Periodic review occurred at or before the defined interval, with a documented outcome.
- Any revision went through change control, with the same preparation, check, and approval sequence as the original.

Filled specimen
The following shows the preparation and approval record completed for an example film-coated tablet, so you can see the level of detail an inspector expects. The company, product, and numbers are illustrative; replace them with your own.
| Field | Entry |
|---|---|
| Product name and strength | Example Tablet 25 mg |
| MPCR document number and version | MPCR-TAB-025-v3 |
| Trigger for this version | Change control CC-2026-071, revised in-process hardness range following a process characterization study |
| Preparer | R. Dlamini, Process Engineering, signed 02 Sep 2026 |
| Independent checker | T. Alvarez, Manufacturing, signed 03 Sep 2026, confirmed hardness range in section 6 matches the approved characterization report |
| QA approval | S. Nakamura, signed 05 Sep 2026, confirmed alignment with the approved application’s process description |
| Effective date | 08 Sep 2026 |
| Prior version withdrawn | Yes, v2 withdrawn from document control issuance system 08 Sep 2026 |
In this example a process characterization study justified a narrower in-process hardness range. The revision went through change control, was independently checked against the underlying study before quality unit approval, and the prior version was withdrawn the same day the new one went effective, so no batch record could be issued from the superseded range.
Common inspection findings this SOP prevents
- A master production record missing the independent-checker signature 211.186 requires, with only a single preparer signature on file.
- Two versions of the same MPCR both appearing “in use” because a superseded version was never formally withdrawn from document control.
- A batch record generated from an outdated MPCR version after a change was approved but before the new version was correctly issued.
- In-process specifications in the MPCR that do not match the specification document or the validated process, discovered only when an investigator compares the two side by side.
- A periodic review requirement that exists in the SOP but has no evidence it was ever performed for a given product.
How to adapt this SOP
- Set your document number, owner, and effective date in the header.
- Point the cross-references in sections 2, 5.4, and 5.6 to your real batch disposition and change control procedures.
- Set the periodic review interval in section 5.5 to match your document control program’s standard cycle.
- If your site uses an electronic batch record (EBR) system, add a step confirming the EBR configuration was updated and verified against the new MPCR version before batch issuance.
- Confirm every regulation in section 7 against the current published version before issue.
