Report: Stability Study Summary and Shelf-Life Justification

A plug-and-play stability study report covering batch and condition results, the ICH Q1E statistical evaluation, significant-change and intermediate-condition findings, the extrapolation basis, and the final shelf-life conclusion, structured to map directly onto CTD Module 3.2.S.7 / 3.2.P.8, with a filled specimen.

Document type: Report

Read and copy the template below into your own quality system. It is a generic starting point for your own internal use, provided as is, with no warranty; see the Terms and License. Adopting it does not by itself create compliance.

This is a ready-to-use stability study report, the document a completed stability protocol produces. It is structured so its sections map directly onto CTD Module 3.2.S.7 (drug substance) or 3.2.P.8 (drug product): the summary and conclusion here becomes .7.1/.8.1, the commitment section becomes .7.2/.8.2, and the data tables feed .7.3/.8.3. Replace every <<FILL: ...>> placeholder with your own specifics. A worked filled specimen follows the template. This document is educational reference, not regulatory or professional advice; confirm every cited regulation against the current published source before you rely on it.

1. Purpose and scope

This report evaluates the stability data generated under <<FILL: protocol number>> for <<FILL: product>> to establish a supported <<FILL: shelf life / retest period>> under the storage condition <<FILL: statement>>, per ICH Q1A(R2) and ICH Q1E.

3. Results by attribute and condition

One row per attribute, summarizing the trend across all batches and time points. Attach full data tables as an appendix.

5. Statistical evaluation (ICH Q1E)

Summarize the outcome of the Q1E shelf-life statistical evaluation worksheet for each attribute that trends over time.

State the conclusion in plain terms, tied directly to the data and statistics above. A conclusion not supported by the confidence-limit method, or an extrapolation exceeding the Q1E caps, is the most common finding on a stability report; read the analysis, then write the sentence, never the reverse.

8. Post-approval stability commitment (maps to CTD 3.2.S.7.2 / 3.2.P.8.2)

10. Acceptance criteria for this report

  • Every attribute’s trend is stated with a conclusion, not only a data table.
  • The statistical evaluation uses the one-sided 95 percent confidence limit, not the mean, and states the poolability outcome.
  • Any extrapolation is checked against both Q1E caps and is consistent with accelerated/intermediate findings.
  • A post-approval stability commitment is stated whenever the data set does not yet fully mature the proposed shelf life.
  • All deviations during the study are listed with their impact on the conclusion, not omitted.

The following shows sections 3, 5, 6, and 7 completed for an illustrative immediate-release tablet, 100 mg, lower assay specification 90.0 percent, 18 months of long-term data on file. Company, product, and numbers are illustrative.

Section 3 extract:

AttributeConditionResult trendSignificant change?Within spec throughout?
AssayLong-term100.1% at 0 months, declining to 97.0% at 18 monthsNoYes
AssayAccelerated99.8% at 0 months, 96.9% at 6 monthsNoYes
Total degradantsLong-term0.1% at 0 months, 0.6% at 18 monthsNoYes

Section 5 extract: Assay modeled linearly; three batches poolable at a 0.25 significance level on both slope and intercept; pooled one-sided 95 percent confidence limit crosses the 90.0 percent lower limit at approximately 50 months.

Section 6 extract: Long-term coverage 18 months. Extrapolation caps: 36 months (double coverage) and 30 months (12 months beyond coverage); binding cap 30 months. Proposed shelf life of 24 months sits inside both caps. Accelerated data showed no significant change through 6 months, supporting extrapolation. Continuation commitment: annual long-term testing continues to 36 months on the same three batches.

Section 7 conclusion (specimen): A 24-month shelf life is proposed for Fictional Tab 100 mg stored at or below 25 C, based on the ICH Q1E one-sided 95 percent confidence limit on pooled assay data from three primary batches, held well inside the extrapolation cap of 30 months, and consistent with accelerated data showing no significant change. No special labeling statement beyond standard room-temperature storage is supported or required.

Common inspection findings this report prevents

  • A report presenting data tables with no stated conclusion, forcing the reviewer to perform the Q1E analysis themselves.
  • A shelf life proposed from the mean trend line rather than the confidence limit.
  • An extrapolation that exceeds the Q1E caps with no flag or discussion.
  • A stability commitment section that is vague or missing when the data set does not yet mature the full proposed shelf life.
  • Deviations during the study omitted from the report instead of listed with their assessed impact.

How to adapt this report

  1. Set your report number and link it to the governing protocol in the header.
  2. Expand section 3 to one row per attribute actually tested; do not compress multiple attributes into one summary row.
  3. If more than one storage condition or climatic zone is being supported, add a sub-table per condition within sections 3 through 6, rolling up to one conclusion in section 7.
  4. Where this report will feed a CTD submission, keep the section numbering and content aligned to 3.2.S.7.1/.2/.3 or 3.2.P.8.1/.2/.3 so the transcription into the dossier is a direct copy, not a re-analysis.
  5. Confirm every regulation in section 11 against the current published version before issue.
