Plan Plug-and-play starting point Clinical & GCP

Plan: Phase-Appropriate IMP Control Strategy

A plug-and-play plan that sets and documents how process validation, specifications, analytical methods, stability, cleaning, equipment qualification, and reprocessing scale across Phase 1, 2, and 3, while naming the controls that never scale down, with acceptance criteria and a filled specimen.

Document type: Plan

Read and copy the template below into your own quality system. It is a generic starting point for your own internal use, provided as is, with no warranty; see the Terms and License. Adopting it does not by itself create compliance.

This is a ready-to-use plan for setting, and defending, the phase-appropriate control strategy for an investigational medicinal product across its clinical development lifecycle. Replace every <<FILL: ...>> placeholder, set your own document numbers, and route it through your normal document control and change management. A filled specimen follows. Verify every cited regulation against the current source before you rely on it. This content is educational reference, not regulatory advice.

1. Purpose

This plan states, for a named investigational product, which controls are fixed at full strength regardless of phase, and which controls scale with process knowledge, so that development, quality, and regulatory functions share one documented control strategy rather than each assuming a different bar. It is the working reference a QP, an inspector, or a new CMC team member reads to understand what is required now and what is planned to tighten, and when.

2. Scope

Covers the drug substance and drug product (or the ATMP equivalent) for <<FILL: product name>> from the current phase through <<FILL: target phase, e.g. end of Phase 2>>. Does not cover commercial process validation planning, which begins under a separate plan once Phase 3 process performance qualification is scoped.

3. The controls that never scale down

State explicitly, for this product, how each fixed control is being met today. These do not get lighter in earlier phases.

4. The controls that scale with phase

Complete this table for the product’s actual trajectory. Each cell should be specific enough that someone could audit against it, not a restatement of the generic phase description.

5. Roles

RoleResponsibility
CMC / development leadOwns this plan, defines and justifies the control-area trajectory, keeps it in step with actual data.
QAConfirms the plan is followed, reviews deviations against it, approves updates.
Qualified Person (EU)Confirms, at certification, that the batch was made under the control strategy this plan currently states.
Regulatory affairsConfirms the IMPD/IND reflects the same control strategy and manages the amendment when it tightens.

6. Acceptance criteria for this plan

  • Every fixed control in Section 3 has a named, current method of being met, not a placeholder.
  • Every scalable control in Section 4 states the trigger that moves it to the next state, not just the current and target states.
  • The plan is reviewed and, where warranted, updated at every phase transition and at every accumulation of data that changes a trigger’s status.
  • The IMPD/IND and the product specification file agree with what this plan states as the current control strategy.

7. Review and update triggers

Review this plan at minimum: at every phase transition, on completion of a method validation that was named as a trigger, on any process or equipment change material enough to affect a row above, and at a fixed interval of <<FILL: e.g. every 12 months>> regardless of other triggers.

Filled specimen

A monoclonal antibody drug product moving from Phase 1 into Phase 2.

Fixed controls: sterility and endotoxin control met via a validated 0.2 micron filtration step and a qualified endotoxin method against the compendial limit; container closure integrity confirmed by a qualified dye-ingress method pending full validation; the manufacturing process is fully recorded in the master batch record though not yet subject to process performance qualification; ALCOA+ enforced through the validated MES and a defined audit trail review schedule; labelling controlled under the site’s IMP labelling SOP with a second check on every run; QP certification performed by the named EU QP before any batch ships.

Scalable controls, selected rows:

Control areaCurrent phase approachTrigger to tightenTarget state at next phase
SpecificationsAssay 90.0-110.0%, limit set from GLP toxicology coverageMethod validated per ICH Q2(R2) and 6 clinical batches on handTighten to 95.0-105.0%, supported by observed range 97-103% across the 6 batches
Analytical methodsPotency assay qualified, not validated; high variability still being characterizedMethod validation protocol executed and passedValidated potency method per ICH Q2(R2), feeding the Phase 2 release spec
Process validationProcess defined and recorded; no PPQPhase 3 entry decision and sufficient process understanding from Phase 2 batchesPPQ campaign scoped and scheduled ahead of Phase 3 manufacture

Review trigger: this plan is scheduled for mandatory review at the Phase 2 to Phase 3 transition decision, and immediately if the potency method validation fails or is materially delayed.

Common inspection findings this plan prevents

  • A control area has clearly tightened in practice (more batches, a newly validated method) but the written plan and the IMPD specification were never updated to match.
  • “Phase-appropriate” is invoked to justify under-controlling a fixed item (sterility, labelling, data integrity) rather than only the scalable ones.
  • No defined trigger exists for when a scalable control should tighten, so tightening happens inconsistently or not at all as data accumulates.

How to adapt this plan

  1. Populate Sections 3 and 4 with your product’s actual current state; do not leave generic phase descriptions in place of your real data.
  2. Set triggers in Section 4 that are objectively checkable (a specific batch count, a specific validation completion), not vague judgment calls.
  3. Keep this plan, the product specification file, and the IMPD/IND specification synchronized at every update; treat a mismatch between them as a finding waiting to happen.
